Abstract: The present invention is directed to advanced genetic engineering techniques for modifying organisms that involve the use of programmable nucleases such as CRISPR/Cas9. The techniques provide a highly efficient and precise method for modifying the genetic material of organisms, allowing for the creation of new varieties with desirable traits. The advanced genetic engineering techniques comprise designing guide RNA sequences specific to the target DNA sequence, introducing the programmable nuclease to the organism, and allowing the nuclease to cleave the DNA at the target site. This induces a double-stranded break that can be repaired through either the non-homologous end joining or homology-directed repair pathway, resulting in the desired genetic modification. The advanced genetic engineering techniques have broad applications in agriculture, medicine, and biotechnology, and have the potential to significantly improve productivity and quality of life.
